What is Journavx (Suzetrigine)?

Suzetrigine is a type of pain signal inhibitor. It targets the NaV1.8 sodium channel in the peripheral nervous system. This is the part of the nervous system that is located outside of the spinal cord and brain. This helps it treat pain in adults. It is not made of opioids like many other pain medications. It avoids the central nervous system—in the brain and spinal cord—and is not made of the same ingredients that commonly cause opioid addiction. The manufacturer responsible for the trials behind this medication is Vertex Pharmaceuticals Incorporated.
Journavx (Suzetrigine) Mechanisms Of Action
Suzetrigine controls pain signaling through its selective inhibition of the NaV1.8 sodium channel. This channel is important to the peripheral nervous system and is considered to be a tetrodotoxin-resistant (TTX-R) isoform. It affects the part of the nervous system that is responsible for detecting and responding to harmful stimuli.
The studies on this medication showed that only about 0.9% of patients had serious adverse effects. Most patients that experienced side effects only experienced mild or moderate effects. The results found the medication to have great potential for being able to fill unmet needs for pain treatment without the use of opioids. It was found to be comparable to HB/APAP without opioid-related risks and gave patients much faster pain relief than the placebo medication it was compared against. It does not cause nausea or fatigue. It also was not found to cause other serious side effects like depression or sedation. Preclinical and clinical trials also found no withdrawal symptoms or drug-seeking behavior resulting from the medication.
Benefits of Journavx (Suzetrigine)
Journavx was officially approved on January 30, 2025. This medication is to be taken every 12 hours and can reduce acute pain significantly 48 hours after surgery. The median time reported for “meaningful” pain relief was 2-4 hours. How long the medication took to kick in varied depending on the type of surgery. Pain relief rated between “good” and “excellent” was experienced by 83% of patients in the phase 3 trial. This medication is only geared toward acute pain management. Studies show it does not work as well for neuropathic and chronic pain management.
It was reported in 2022 that the opioid crisis alone is responsible for hundreds of thousands of deaths and many more addictions. NSAIDS were all that were available before Journavx besides some anti-convulsant medications and antidepressants that could potentially help with pain relief. These medications can cause serious side effects like euphoria and slowed breathing. They also cause side effects such as nausea and drowsiness. These medications are very easy to become addicted to and a tolerance to these medications can be built fast. Journavx is non-addictive and has much less severe side effects.
Safety and Side Effects of Journavx (Suzetrigine)
Clinical trials did identify some possible side effects for those taking Journavx. About 2.1% of patients experienced itching and about 1.3% of patients experienced muscle spasms. About 0.5% experienced rash or Creatine Phosphokinase (CPK) increased blood levels. Only 1% reported nausea or vomiting in the first couple of trials.
This medication should not be used by those with liver issues or those using CYP3A inhibitors, inducers, or substrates. This could worsen side effects or cause a bad reaction. It should also not be used with hormonal birth control. Studies have yet to determine if this medication is safe to use while breastfeeding or pregnant. This medication is still being tested even though it has already been approved. These studies will continue to look into the long-term safety of using the medication and its impact over time.
Journavx (Suzetrigine) Compared To Current Pain Treatments
Journavx is different from other existing pain management medications. Gabapentin and pregabalin are other non-opioid medications that have been used to treat pain. These medications are a part of the anti-convulsant medication class and also impact nerve function. They interact with the α2-δ-1 and α2-δ-2 subunits of calcium channels in neurons. They minimize the release of neurotransmitters involved in pain signaling. Journavx is also different than traditional opioid treatments that bind to the spine and brain to minimize the feelings of pain. It is much less addictive than opioids and does not cause as many side effects as gabapentin and pregabalin or opioid medications. Gabapentin and pregabalin are usually used for neuropathic pain. Journavx is mostly used to treat pain after surgery.
Journavx has contraindications with CYP3A inhibitors and interacts with hormonal birth control. This may limit its use for some patients. There is not enough information yet to see how this medication impacts children. This medication may also be expensive for users. Single 50-milligram pills are being listed at $15.50 and need to be taken twice per day. This medication costs hundreds of dollars each week without insurance coverage.
